Longevity for Dogs: The Science of Healthy Canine Aging

A note on language. We use structure/function language throughout. Why mitochondria matter for canine healthspan
Mitochondria are the small organelles inside almost every cell that convert nutrients into ATP — the chemical energy that powers everything from muscle contraction to immune-cell signaling. Across mammalian species, mitochondrial decline is one of the most consistent features of aging. Dysfunctional mitochondria produce less ATP per nutrient unit and more oxidative byproducts; their accumulation is associated with loss of muscle mass, reduced exercise tolerance, and a range of age-related functional declines.
The dog-specific aging literature is less developed than the human or mouse literature, but the underlying biology is conserved. The general framing — mitochondria as nutritional targets to maintain muscle health and physical function during aging — is laid out in a 2024 Sports Medicine review 1. The broader geroscience framework, including the "hallmarks of aging" that include mitochondrial dysfunction, has been synthesized in a 2025 Cell paper on precision geromedicine 2 and in canonical caloric-restriction-mimetic reviews 34.
What does mitochondrial decline look like in an aging dog in practice? Slower stair-climbing, shorter walks, reduced muscle mass on the topline (sarcopenia-like changes), longer recovery after activity, and reduced enthusiasm for the things that used to be easy. None of these are diagnostic on their own; they are general patterns. The Vector Canis premise: support the mitochondrial-quality machinery while the dog is still healthy, rather than waiting for problems.
Urolithin A — mitophagy activator
Urolithin A is the gut-bacterial metabolite that humans (and many dogs) produce when ellagitannins from pomegranate, walnut, and certain berries reach the gut microbiome. The conversion is microbiome-dependent — not every individual produces urolithin A efficiently from dietary precursors. A 2022 review covers the gut-microbial-metabolite biology and biological activities of urolithins 5. Direct supplementation bypasses the microbiome conversion.
Mechanistically, urolithin A activates mitophagy — the selective autophagy of damaged mitochondria. The pivotal mechanism paper, Ryu et al. 2016 in Nature Medicine, showed mitophagy induction and lifespan prolongation in C. elegans alongside increased muscle function in rodents 6. Subsequent reviews lay out the broader urolithin A health story 789.
The human clinical evidence is substantial. Andreux 2019 in Nature Metabolism established safety and a mitochondrial-health molecular signature in older adults at 250–1000 mg/day for 28 days 10. Liu 2022 in JAMA Network Open showed muscle-endurance improvements in a four-month RCT 11. Singh 2022 in Cell Reports Medicine showed muscle-strength and exercise-performance improvements in middle-aged adults at 500 mg/day 12. A 2024 systematic review summarized the human data 8.
The honest translational framing: the strongest urolithin A clinical data is in humans; the mechanism work spans species; canine-specific RCTs at Vector Canis doses do not yet exist in the public literature. We body-weight-scale from the human-trial dose range. We want to be honest about that — most companion-animal longevity research follows this same translational pattern.
Calcium alpha-ketoglutarate (Ca-AKG)
Alpha-ketoglutarate is a Krebs-cycle intermediate. The Krebs cycle is the central metabolic pathway by which cells extract energy from glucose, fatty acids, and amino acids; AKG sits at a hub position. Calcium alpha-ketoglutarate is the supplemental calcium salt of AKG.
The mouse data is striking. A 2020 study in Cell Metabolism reported that AKG, administered late in life, extended median lifespan and compressed morbidity in aging mice 13. The mechanistic story has roots in earlier work: a 2014 Nature paper showed that AKG extends lifespan in C. elegans by inhibiting ATP synthase and TOR signaling 14. A 2020 Nature Communications paper described AKG's effects on age-related osteoporosis in mice via regulation of histone methylations 15. A 2022 review surveyed AKG dietary supplementation in humans and laid out the open questions 16.
Canine-specific Ca-AKG dose-response data is essentially absent in the published literature. We body-weight-scale from the rodent food-supplementation doses, which translate to roughly 1–2 g/day in adult humans. Vector Canis doses Ca-AKG at a level appropriate for adult-to-senior medium-large breeds with smaller-breed dose guidance on the label.
Spermidine
Spermidine is a polyamine — a small organic molecule involved in many cellular processes — that is studied primarily for its role in inducing autophagy. The 2018 Science review by Madeo et al. is the standard reference on spermidine in health and disease 17; the same group's 2019 Cell Metabolism review framed spermidine and related polyamines as part of the caloric-restriction-mimetic class 3.
For cardiovascular healthspan specifically, a 2018 Circulation Research paper covered autophagy's role in cardiovascular aging 4. Autophagy induction is one of the cleaner mechanism stories in the longevity-supplement space: the mechanism is conserved across species, the safety profile is established, and the mode of action (recycling damaged cellular components) is unambiguous in cell biology.
Canine-specific spermidine data is thin. We include spermidine in Vector Canis because the autophagy-induction mechanism is conserved across mammals and the safety profile is well-established.
Resveratrol — a complementary polyphenol
Resveratrol is a stilbenoid found in red grape skins. It has been studied for its effects on cellular-energy pathways (including SIRT1 activation in some contexts) and as a complementary polyphenol in stacks targeting mitochondrial function. The human and animal literature is broad; resveratrol's primary role in Vector Canis is as a complementary low-dose polyphenol to support the mitochondria-and-autophagy story established by urolithin A, Ca-AKG, and spermidine.
A note on fisetin
NK9 includes fisetin at a low dose; Vector Canis does not include fisetin in the primary formula. The reason: fisetin is studied primarily for its senolytic mechanism (clearing senescent cells) 1819; we put it in the immune-support formula where the mechanism story is most direct (chronic low-grade inflammation reduction). The Vector Canis longevity rationale prioritizes mitophagy, Krebs-cycle support, and autophagy. Senolytic stacking remains a valid complementary approach; some owners may want to combine NK9 (which has fisetin) with Vector Canis.
Joint supplements — a complementary not-this category
The canine osteoarthritis-supplement literature is the most-developed segment of the canine supplement field. A 2017 RCT of a curcuminoids-containing diet supplement reported improvement on standard osteoarthritis assessments 20; a 2022 study evaluated green-lipped mussel + curcumin 21; a 2016 review surveyed the broader joint-supplement evidence base 22. A 2006 chapter and 2011 review covered nutrition and immune function in companion animals more broadly 2324.
Vector Canis is not a joint supplement. If your dog has osteoarthritis or joint comfort concerns, the joint-supplement evidence base is the right place to start; Vector Canis is a complement, not a substitute.
How to think about supplement-stacking for adult and senior dogs
Foundational interventions first. Maintaining a lean body condition, age-appropriate exercise, and the right diet for breed and age account for most of the variance in canine healthspan. A daily longevity supplement is a layer above these foundations, not a substitute.
Above that, Vector Canis is the mitochondria-and-autophagy stack; NK9 is the immune-and-inflammation stack. Joint supplements (if needed) are the connective-tissue stack. The three target different mechanisms and can be combined under veterinary guidance.
What the research does not say
Honest limits:
- The mouse Ca-AKG lifespan-extension data is striking 13; canine-specific lifespan-extension data does not exist. Dose translation is approximate, not validated.
- Urolithin A's strongest clinical data is in humans, not dogs. We body-weight-scale; we do not have a published canine RCT at Vector Canis doses.
- Spermidine and resveratrol mechanism work is broad and cross-species; the canine-specific RCT base is thin.
- Mitochondrial-function biomarker improvements in human RCTs (Liu 2022, Singh 2022) are real signals; whether they translate to canine functional outcomes at body-weight-scaled doses is an open question.
- These are dietary supplements, not pharmaceuticals. We support cellular and tissue functions in dogs; we do not make therapeutic claims.

- How is Vector Canis different from joint or hip supplements?
- Joint supplements (glucosamine, chondroitin, MSM, green-lipped mussel, sometimes curcumin) target connective-tissue and joint-comfort endpoints, which is well-studied in dogs with osteoarthritis [cit_comblain_2016_supplements_dog_oa_review]. Vector Canis targets a different axis — mitochondrial quality control via mitophagy and Krebs-cycle metabolism. The two are complementary, not interchangeable.
- Is Vector Canis safe for healthy adult dogs?

- Should I combine Vector Canis with NK9?
- Vector Canis and NK9 target different axes — longevity-mechanism support and immune-mechanism support. They are compatible. If you are considering stacking, talk to your veterinarian about combined dosing and any specific health considerations for your dog.
- What age should I start Vector Canis?
- Most owners start in mid-adulthood — roughly age 5–7 for medium-large breeds, age 7–9 for small breeds — because mitochondrial decline is gradual and earlier support is the relevant frame. For seniors already showing reduced energy or muscle mass, mitochondria-targeted support is the right time to start regardless of age. The honest framing: there is no single 'right' age.
